SFN (Stratifin), also called 14-3-3 Sigma, is a p53-induced tumor suppressor gene which is activated in response to DNA damage, causing cell cycle arrest at G2 phase by blocking cdc2-cyclin B1 complex from entering the nucleus. It is inactivated in breast, lung, prostate, liver and gastric cancer. It is associated with poor prognosis when its down-regulation is observed in epithelial ovarian cancer. SFN expression could contribute to cancer cell proliferation and the development and/or progression of human gastrointestinal cancer. Protein function: Adapter protein implicated in the regulation of a large spectrum of both general and specialized signaling pathways. Binds to a large number of partners, usually by recognition of a phosphoserine or phosphothreonine motif. Binding generally results in the modulation of the activity of the binding partner. When bound to KRT17, regulates protein synthesis and epithelial cell growth by stimulating Akt/mTOR pathway. May also regulate MDM2 autoubiquitination and degradation and thereby activate p53/TP53.
