Product information "Anti-Sphingolipid Receptor (Edg8/S1P5) (APC)"
Reagents are stable for twelve months from the date of EDG-8 is also known as sphingosine 1-phosphate receptor-5 (S1P5) and nerve growth factor-related G protein-coupled receptor-1 (NRG-1). EDG-8 is a 398aa seven transmembrane receptor that binds the lysolipid phosphoric acid mediator, sphingosine 1-phosphate. Extracellular portions of human EDG-8 show 96% and 97% aa identity with mouse and rat EDG-8, respectively. Isoform 1 is expressed at a low level in peripheral tissues. Isoform 2 has an alternate C- terminal that is 88aa shorter and is expressed mainly in brain, spleen, and peripheral blood mononuclear cells. EDG-8 is upregulated in large granular lymphocytic leukemias. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilution: Flow Cytometry: 10ul labels, 1x10e6 cells. Validated using HEK293 human embryonic kidney cell line transfected with human and mouse EDG8 and eGFP., Optimal dilutions to be determined by the researcher. Hybridoma: Mouse myeloma cells with B cells from a mouse immunized with BaF3 cells transfected with recombinant human endothelial differentiation G protein-coupled receptor-8. Storage and Stability: Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: APC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
