p53 tumor suppressor gene mutations are one of the most common genetic alterations in human tumors, including lymphomas. Altering or inactivating p53 by mutation or reaction with oncogenes or viruses may cause depletion of p53, or conversion of p53 from a tumor suppressor gene to a transforming oncogene. The nuclear protein p53 is associated with cell transformation and proliferation, with definitive possibilities as a marker in malignant lymphoma progression. p53's role in the multistep process leading to cancer is still being investigated, with an emphasis on its relation and control over additional tumor suppressors. Applications: Suitable for use in Western Blot, Immunoprecipitation, Immunohistochemistry, ELISA and FACS analysis. , Other applications not tested. Recommended Dilution: Western Blot: 1:1000-1:2000. , Immunohistochemistry (Frozen): 1:250-1:500, Immunohistochemistry (Paraffin): 1:250-1:500, antigen retrieval is not essential but may optimize staining., Immunoprecipitation: 10ug/mg protein lysate. By IP this antibody reacts with only mutant p53 protein under non-denaturing condiions., Optimal dilutions to be determined by the researcher. Positive Control: MDA-MB-231 cell line, Storage and Stability: May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile 40-50% glycerol, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.,
