Product information "Anti-Neuropeptide Y Y2 Receptor (NPY Y2R)"
Several subtypes of NPY Receptors have been described of which the cloned Y1-, Y2- and Y5-receptors, members of the superfamily of G-protein coupled receptors, appear to be the endogenous binding sites for NPY. The Y(2) receptor is the predominant Neuropeptide Y (NPY) receptor subtype in the brain. Y(2) receptor-mediated effects have been reported principally based on the observations using the C-terminal fragments of NPY. Recent studies indicate an involvement of the receptor in food intake, gastrointestinal motility, cardiovascular regulation, and neuronal excitability. Applications: Suitable for use in ELISA, Immunohistochemistry and Immunofluorescence. Other applications not tested. Recommended Dilution: Immunohistochemistry: 1:500-1:1000, Used on perfusion fixed tissue. Perfusion: 1) calcium-free Tyrode's solution, 2) paraformaldehyde-picric acid fixative and 3) 10% sucrose in PBS. Tissues were dissected and stored overnight in 10% sucrose, PBS., Immunofluorescence (Indirect): Incubate slides with blocking buffer for 1 hour at RT. Dilute N2176-63G with blocking buffer. Remove blocking buffer from slide and incubate with N2176-63G for 18-24 hours at 4°C. Rinse slides 3X and then incubate with appropriate secondary antibody for 1 hour at RT. Rinse slides again 3X and set coverslip. Examine staining using fluorescence microscopy., Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
