Anti-CD40 (Biotin)

Human CD40 is a member of the tumor necrosis factor (TNF) receptor family and is present on all B cells except plasma cells. CD40 is also found on some epithelial cells, carcinomas and lymphoid dendritic cells. CD40 plays an important role in B cell activation and the interaction with its ligand CD154 (CD40 Ligand) is essential for isotype switching (1). The CD40-CD154 interaction appears to have a role in artherosclerosis and cardiovascular disorders (3,4,5). Applications: Suitable for use in Flow Cytometry, ELISA and Immunoprecipitation. Other applications not tested. Recommended Dilutions: Flow Cytometry: 10ug/ml. 80ul labels 5x10e5 cells., Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. 5x10e5 cultured Raji cells were washed and incubated 45 minutes on ice with 80ul of C2392-02B at a concentration of 10ug/ml. , 2. Cells were washed twice and incubated with 2º reagent Streptavidin/R-Phycoerythrin (S7973-88P), after which they were washed three times, fixed and analyzed by FACS. , 3. Cells stained positive with a mean shift of 2.37 log10 fluorescent units when compared to a Mouse IgG1/Biotin negative control at a similar concentration. , 4. Binding was blocked when cells were pre-incubated 10 minutes with 20ul of 0.5mg/ml anti-CD40 antibody (C2392-02A). Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
