Interleukin-5 (IL5), also known as eosinophil differentiation factor (EDF), is a lineage-specific cytokine for eosinophilpoiesis . It regulates eosinophil growth and activation, and thus plays an important role in diseases associated with increased levels of eosinophils, including asthma. IL5 has a similar overall fold to other cytokines (e.g., IL2, IL4 and GCSF), but while these exist as monomeric structures, IL5 is a homodimer. The fold contains an anti-parallel 4-alpha-helix bundle with a left handed twist, connected by a 2-stranded anti-parallel beta-sheet . The monomers are held together by 2 interchain disulfide bonds. Description: Recombinant Rat IL-5 produced in E.Coli is a dimeric, non-glycosylated polypeptide chain containing 113 amino acids and having a molecular mass of 13074 Dalton. The Recombinant Rat IL-5 is purified by proprietary chromatographic techniques. Amino Acid Sequence: The sequence of the first five N-terminal amino acids was determined and was found to be Met-Glu-Ile-Pro-Met, Dimers and Aggregates: Less than 1% as determined by silver-stained SDS-PAGE gel analysis. Biological Activity: Rat IL-5 is fully biologically active when compared to standard. The ED50 as determined by the dose-dependent stimulation of the proliferation of BCL-1 cells was found to be < 0.15ng/ml. Endotoxin: < 0.1ng/ug (IEU/ug) of Recombinant Rat IL-5. Protein Content: Protein quantitation was carried out by two independent methods:, 1. UV spectroscopy at 280 nm using the absorbency value of 0.635 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics). , 2. Analysis by RP-HPLC, using a standard solution of IL-5 as a Reference Standard. Gene: Name: Il5, Synonyms: Il-5, Solubility: It is recommended to reconstitute the lyophilized Rat IL-5 in sterile 18M -cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions. Stability: Lyophilized Rat IL-5 although stable at room temperature for 3 weeks, should be stored desiccated below -180C. Upon reconstitution Rat IL-5 should be stored at 40C between 2-7 days and for future use below -180C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please avoid freeze-thaw cycles.
