Poly [ADP-ribose] polymerase 1 (PARP1), also known as ADPRT or PPOL is an enzyme that in humans is encoded by the PARP1 gene. PARP1 gene is mapped to 1q42.12. This gene encodes a chromatin-associated enzyme, poly(ADP-ribosyl)transferase, which modifies various nuclear proteins by poly(ADP-ribosyl)ation. The modification is dependent on DNA and is involved in the regulation of various important cellular processes such as differentiation, proliferation, and tumor transformation and also in the regulation of the molecular events involved in the recovery of cell from DNA damage. In addition, this enzyme may be the site of mutation in Fanconi anemia, and may participate in the pathophysiology of type I diabetes. Applications: Suitable for use in Western Blot, Immunohistochemistry and Immunocytochemistry. Other applications not tested. Recommended Dilutions: Western Blot: 0.1-0.5ug/ml. The detection limit is ~0.25ng/lane under reducing conditions., Immunohistochemistry (Paraffin): 0.5-1ug/ml. Boiling the paraffin sections in 10mM citrate buffer, pH 6.0, for 20 minutes is required for the staining of formalin/paraffin sections., Immunocytochemistry: 0.5-1ug/ml, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
