PHD1 catalyzes the post-translational formation of 4-hydroxyproline in hypoxia-inducible factor (HIF) alpha proteins. Hydroxylates HIF-1 alpha at 'Pro-402' and 'Pro-564', and HIF-2 alpha. It functions as a cellular oxygen sensor and, under normoxic conditions, targets HIF through the hydroxylation for proteasomal degradation via the von Hippel-Lindau ubiquitination complex. It may play a role in cell growth regulation. Applications: Suitable for use in Immunohistochemistry, ELISA and Western Blot. Other applications not tested. Recommended Dilutions: Immunohistochemistry: 1:50-1:100. Formalin-fixed and paraffin-embedded human testis tissue reacted with P3373-05E, which was peroxidase-conjugated to the secondary antibody, followed by DAB staining. , ELISA: 1:1000, Western Blot: 1:100-1:500, Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
