The p53 gene is located on human chromosome 17p13.1. Mutation and/or allelic loss of this gene is one of the causes of the occurrence of a variety of mesenchymal and epithelial tumors. If it occurs in the germ line, such tumors run in families. p53 binds to a DNA consensus sequence, the p53 response element, and it regulates normal cell growth cycle events by activating transcription of genes, involved either in progression through the cycle, or causing arrest in G1 when the genome is damaged. In most transformed and tumor cells the concentration of p53 is increased 5-1000 fold over the minute concentrations (1000 molecules cell) in normal cells, principally due to the increased half-life (4 h) compared to that of the wild-type (20 min). p53 localizes in the nucleus, but is detectable at the plasma membrane during mitosis and when certain mutations modulate cytoplasmic/nuclear distribution. p53 binds damaged DNA non-specifically via its C-terminal 75 amino acids (1) which in turn may be responsible for subsequent activation, sequence specific DNA binding and WAF-1 induction allowing DNA repair to take place (2,3). p53 downregulates bcl-2 expression and upregulates bax expression, but may not always be necessary for apoptosis (4,5)., , Applications: , Western Blot: 1-10ug/ml , Immunoprecipitation , Immunohistochemistry , Optimal dilutions to be determined by researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
