TBK, also known as IkappaB kinase (IKK)-related kinase NAK, belong to the serine/threonine IKK kinase family and is thought to play a role in NF-kappa-B (NF-kappaB) activation. The NF-kappaB complex of proteins is inhibited by I-kappa-B (IKB) proteins, which inactivate NF-kappaB by trapping it in the cytoplasm. Phosphorylation of serine residues on the IKB proteins by IKB kinases marks them for destruction via the ubiquitination pathway, thereby allowing activation and nuclear translocation of the NFKB complex. The protein encoded by this gene is similar to IKB kinases and can mediate NF-kappaB activation in response to certain growth factors. For example, the protein can form a complex with the IKB protein TANK and TRAF2 and release the NF-kappaB inhibition caused by TANK. Applications: Suitable for use in Western Blot and Immunoprecipitation. Other applications not tested. Recommended Dilution: Western Blot: 1:500-1:2000, Optimal dilutions to be determined by the researcher. Positive Control: RIPA lysates from HEK293 cells. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
