Product information "Anti-Growth and Differentiation Factor 5 (GDF5)"
Growth Differentiation Factor 5 (GDF5) is a member of the bone morphogenetic protein (BMP) subfamily of the TGF-beta superfamily. It is synthesized as a large precursor protein that consists of an N-terminal 19aa signal sequence, a 362aa pro region and a 130aa C-terminal mature peptide (aa376-495). Homodimeric GDF5 activity is mediated by receptor complexes containing type I (BMPR-IB) and type II (BMPR-II or Activin R-II) serine/threonine kinase receptors. GDF5 has multiple functions including the regulation of myogenesis, chondrogenesis, bone morphogenesis and neuronal differentiation and survival. Applications: Suitable for use in ELISA and Neutralization. Other applications have not been tested. Recommended Dilutions: Neutralization (ND50): 2-10ug/ml in the presence of 3ug/ml of recombinant mouse GDF5, using the ATDC5 cell line., Definition: Measured by its ability to neutralize GDF?5-induced alkaline phosphatase production in the ATDC5 mouse chondrogenic cell line., Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
