Granzymes are neutral serine proteases, which are stored in specialized lytic granules of cytotoxic T lymphocytes (CTLs) and in natural killer (NK) cells. A number of granzymes A to G, have been isolated and cloned from mouse CTLs and NK cells, however in man, fewer have been cloned and identified. One of these, human granzyme B, is involved in target cell apoptosis during lymphocyte mediated cytotoxicity. Exocytosis of granzyme containing granules in the cytoplasm of the target cell leads to the induction of DNA fragmentation and apoptosis. Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Immunohistochemistry (Paraffin): 1:50-1:200. Requires antigen retrieval using heat treatment prior to staining of paraffin sections. Sodium citrate buffer pH 6.0 is recommended., Optimal dilutions to be determined by the researcher. Positive Control: Granzyme B, is expressed within the granules of cytolytic lymphocytes (CTLs). Hybridoma: Sp2/0 myeloma cells with spleen cells from Balb/c mice. Recommended Secondary Reagents: I1904-65W IgG, F(ab')2 (HRP) Pab Rb xMo, I1904-66C IgG, F(ab')2, X-Adsorbed (HRP) Pab Rb xMo, Storage and Stability: May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile 40-50% glycerol, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
