Granzymes are exogenous serine proteases that are stored in the cytotoxic granules of activated T cells and NK cells. Upon target cell contact, the contents of these granules are directionally exocytosed and, with the assistance of perforin, the granzymes enter the cytosol of the target cell. In the target cell, granzyme B rapidly induces apoptosis by activating both caspase-mediated as well as caspase-independent pathways. Apart from the storage granules of cytotoxic cells granzymes can also be detected in biological fluids such as serum and plasma. Elevated levels of soluble granzymes have been found in the sera of patients with increased cytotoxic responses, caused by various viral infections, as well as in the synovial fluid of rheumatic arthritis patients. A high percentage of GrB positive CTL has also been found in the glands of patients suffering from Hodgkin`s disease and in CTL infiltrates in transplant rejection. Applications: Suitable for use in ELISA and Flow Cytometry. Other applications not tested. Recommended Dilutions: ELISA: 1:200-1:400, Flow Cytometry: 1.2ug per 10e6 cells, Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
