Fibroblast growth factors (FGFs) produce mitogenic and angiogenic effects in target cells by signaling through cell surface receptor tyrosine kinases. There are four members of the FGF receptor family: FGFR-1 (flg), FGFR-2 (bek, KGFR), FGFR-3 and FGFR-4. Each receptor contains an extracellular ligand binding domain, a trans- membrane domain and a cytoplasmic kinase domain. Following ligand binding and dimerization, the receptors are phosphorylated at specific tyrosine residues. Seven tyrosine residues in the cytoplasmic tail of FGFR-1 can be phosphorylated: Tyr463, Tyr583, Tyr585, Tyr653, Tyr654, Tyr730 and Tyr766. Tyrosines 653 and 654 are important for catalytic activity of activated FGFR and are essential for signaling. The other phosphorylated tyrosine residues may provide docking sites for downstream signaling components such as Crk and PLCgamma. Activating mutations within fibroblast growth factor receptor 3 (FGFR-3) are responsible for human skeletal dysplasias including achondroplasia and the neonatal lethal syndromes thanatophoric dysplasia types I and II. Several of these same FGFR-3 mutations as well as overexpression of FGFR- 3 proteins have also been identified somatically in human cancers, including multiple myeloma, bladder carcinoma and cervical cancer. Thus, FGFR-3 may represent a potential target for therapy. Applications: Suitable for use in Western Blot, Immunoprecipitation, Immunohistochemistry and Immunofluorescence/, Immunocytochemistry. Other applications not tested. Recommended Dilutions: Western Blot: 1:1000, incubate membrane with diluted antibody in 5% BSA, 1X TBS, 0.1% Tween-20 at 4°C with gentle shaking, overnight., Immunoprecipitation: 1:50, Immunohistochemistry (Paraffin): 1:50. Antigen retrieval with citrate buffer required., Immunofluorescence (IC): 1:200, Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
