Cartilage Oligomeric Matrix Protein (COMP), also known as Thrombospondin-5, is a 110kD modular matricellular protein. COMP contains a non-collagenous coiled-coil domain, four EGF-like repeats, seven TSP type-3 repeats, and a globular TSP C-terminal domain. The coiled coil domain mediates the association of COMP into disulfide-linked homopentamers. An RGD sequence in the third TSP type-3 repeat mediates chondrocyte attachment via integrins alpha5beta1 and alphaVbeta3. COMP is upregulated in inflammation and some carcinomas. , , Applications:, Suitable for use in Western Blot and Immunoprecipitation. Other applications not tested. Recommended Dilution: Western Blot: 2ug/ml detects human COMP in cell or tissue extracts., Immunoprecipitation: 25ug/ml immunoprecipitates recombinant human COMP from conditioned cell culture media. , Optimal dilutions to be determined by the researcher. Western Blot Buffers: Blotting Buffer: 25mM Tris, pH 7.4, 0.15M sodium chloride, 0.05% Tween-20, Blocking Solution: 5% nonfat dry milk in Blotting Buffer, Antibody Solution: 5% nonfat dry milk in Blotting Buffer, Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
