CD117 or c-kit, a transmembrane receptor tyrosine kinase, is a receptor for a growth factor encoded by the steel locus and known as steel factor (SLF), stem cell factor (SCF), mast cell growth factor MGF or KIT Ligand (KL). The binding of the stem cell factor (SCF) to the c-kit-encoded receptor tyrosine kinase (Type III) stimulates a variety of biochemical responses that culminate in cellular proliferation, migration or survival. c-kit plays an important role in hematopoiesis, melanogenesis and gametogenesis. The antigen is expressed in many tissues and cells: 0.6-3.4% of normal bone marrow cells which comprise BFU-E, CFU-GEMM, CFU-GM and CFU-G. 19-51% of CD34+ bone marrow cells co-express c-kit. c-kit/SCF is involved in development of several lineages of stem cells, such as germ cells, neural crest-derived melanocytes, and hematopoietic precursor cells. Applications: Suitable for use in Immunohistochemistry, Flow Cytometry, Immunoprecipitation, Western Blot, and blocking of ligand binding. Other applications not tested. Recommended Dilutions: Immunohistochemistry: Frozen tissue, Optimal dilutions to be determined by the researcher. Positive Controls: Skin tissue, normal breast epithelium, melanocytes, mast cells and glial cells. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
