Cadherins are transmembrane glycoproteins involved in Ca++-dependent cell adhesion. The cadherin family, including E-Cadherin, N-Cadherin, P-Cadherin and R-Cadherin, plays an important role in tissue morphogenesis and structure. E- cadherin is a 120kD protein that mediates Ca++-dependent adhesion between epithelial cells and is involved in the generation of epithelial layers during development and the and maintenance of epithelial tissue in adults. The presence of E-cadherin has been reported in other cell types, including osteoblasts, peripheral nerve Schwann cells and Langerhans cells. The large number of papers linking E-cadherin, or lack of E-cadherin expression, with a broad range of cancers suggests that its immunodetection in tissue may be useful in the diagnosis and prognosis of a variety of cancers. Tumor analysis by immunohistochemical techniques revealed that the reduction and/or loss of E-cadherin expression in carcinomas (including tumors of the neck, prostate, esophagus and other cancers) correlates positively with tumor cell invasion and metastasis. Studies have also associated reduced expression of E-cadherin with invasiveness and poor prognosis in breast cancer and poor prognosis in bladder cancer. Other studies have indicated that analysis of E-cadherin and N-cadherin expression may be useful in differentiating pleural mesotheliomas (N-cadherin positive) and lung adenocarcinomas (E-cadherin positive). Applications: Suitable for use in Immunohistochemistry. Other applications not tested. Recommended Dilutions: Immunohistochemistry (Formalin-fixed, paraffin-embedded or frozen sections): 1:50-1:200. Epitope retrieval required (Citrate Buffer, pH 6)., Optimal dilutions to be determined by the researcher., ., Recommended Positive Control: , Non-neural epithelia tissue , Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
