BMI1 is a 326aa containing protooncogene belonging to the polycomb gene family with a RING-type zinc finger domain. A transcription factor and a senescence regulator, BMI1 is a component of the Polycomb group (PcG) multiprotein PRC1 complex, required to maintain the transcriptionally repressive state of many genes, including Hox genes, throughout development. An important role of BMI1 has been reported in regulating proliferation of stem cells and tumor cells in diverse adult epithelial tissues involved in human colorectal carcinogenesis by repressing the INK4a:ARF proteins (CDKN2A). This protein also plays an essential role in the generation of self-renewing adult hematopoietic stem cells and in the maintenance and expansion of immature granule cell precursors. BMI1 cooperates with the MYC oncogene to produce B-lymphomas. Ubiquitous expression is seen in most of the tissues. Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 2-3ug/ml, Immunohistochemistry (paraffin): 5ug/ml, Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
