Water is a critical component of all living cells. Interestingly, tissue membranes show a great degree of water permeability. Mammalian red cells, renal proximal tubules, and descending thin limb of Henle are extraordinarily permeable to water. Water crosses hydrophobic plasma membranes either by simple diffusion or through a facilitative transport mechanism mediated by special protein "aquaporin". Aquaporin, are integral membrane proteins from a larger family of major intrinsic proteins (MIP) that form pores in the membrane of biological cells. Over the last decade, genes for several members of the aquaporin family have been cloned, expressed, and their distribution studied in many tissues. Aquaporin 1 (APQ1) is a 28kD integral membrane protein which was originally identified in red blood cells and renal proximal tubules. AQP1 is also expressed by the choroid plexus and various other tissues. Applications: Suitable for use in ELISA, Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilutions: ELISA: 0.5-1.0ug/ml, Western Blot: 0.5-10ug/ml using Chemiluminescence technique. Reveal two bands, a lower 28kD band corresponding to unglycosylated form and an upper high molecular 35-60kD glycosylated component (Denker et al. 1988, Smith & Agre 1991). We recommend the use of 0.5-1.0% milk in all primary/secondary antibody-enzyme conjugate incubations in order to suppress non-specific bands. , Immunohistochemistry: 2-10ug/ml on paraformaldehyde fixed tissue sections. , Optimal dilution determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
