FMS-related tyrosine kinase 3 (FLT3, also called Flk2), a member of the type III receptor tyrosine kinase family that includes c-Kit, PDGF and M-CSF receptors, is expressed on early hematopoietic progenitor cells, and supports growth and differentiation within the hematopoietic system. FLT3 is activated after binding with its ligand FL, which results in a cascade of tyrosine autophosphorylation and tyrosine phosphorylation of downstream cellular targets. The p85 subunit of PI3 kinase, SHP2, GRB2 and Shc are associated with FLT3 after FL stimulation. Tyrosine 591 is located in the juxtamembrane region of FLT3. It may play an important role in regulation of FLT3 tyrosine kinase activity. Somatic mutations of FLT3 consisting of internal tandem duplications (ITDs) occur in 20% of patients with acute myeloid leukemia. Human Flt3 (GenBank Accession No. NM_004119), (a.a. 564-end) with N-terminal His tag, , MW 54.2kD, expressed in an Baculovirus infected Sf9 cell expression system. Specific Activity: 225pmol/min/ug , Assay Conditions:, Enzyme reaction is conducted in a buffer containing 50mM HEPES, pH7.5, 10mM magnesium chloride, 1mM EDTA, 200uM ATP, 0.01% Brij-35 and 2uM substrate (Tyr peptide 2) at RT for 1 hour. Applications: Suitable for use in the study of enzyme kinetics, screening inhibitors and selectivity profiling. Other applications not tested. Recommended Dilutions: Optimal dilutions to be determined by the researcher. Storage and Stability: Aliquot to avoid repeated freezing and thawing and store at -70°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Aliquots are stable for at least 6 months.
