In the classical pathway of NF-kappaB activation, phosphorylation of the inhibitor of NF-kappaB (IkappaBalpha) releases the inhibitor from NF-kappaB, allowing IkappaBalpha degradation and NF-kappaB activation and nuclear import. BAY 11-7085 is an irreversible inhibitor of IkappaBalpha phosphorylation, preventing activation of NF-kappaB by cytokines and lipopolysaccharide (IC50 = 10 µM). It blocks gene expression that is regulated through the classical pathway of NF-kappaB activation and in this way blocks apoptosis, cell adhesion, and inflammation.
