PGC-1a is a 97-120kD member of the PGC-1 family of proteins. It is expressed in select cell types, including brown adipocytes, skeletal muscle and hepatocytes. PGC-1a participates in both RNA processing and transcriptional coactivation in conjunction with multiple nuclear hormone receptors such as PPARg, RAR and TR. Human PCG-1a is 798aa in length. It contains an LxxLL nuclear receptor binding motif aa144-148, one PPAR-g interaction domain aa293-339, two NLSs and an RNA binding/processing region aa566-710. PGC-1a activity is regulated by phosphorylation. AMPK is known to phosphorylate Thr178 and Ser539, promoting cotranscriptional activity. Conversely, Akt-mediated phosphorylation at Ser571 is reported to downregulate PGC-1a activity. This latter effect is achieved by an initial Ser571 phosphorylation, followed by GCN5 binding and subsequent PCG-1a acetylation that promotes PGC-1a dissociation from target gene promoters. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilutions: Western Blot: 1ug/ml using HeLa human cervical epithelial carcinoma cell line and Jurkat human acute T cell leukemia cell line treated with recombinant human IGF-1, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months after receipt at -20°C. Reconstitute with PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
