Product information "Anti-phospho-Met, (Tyr1234/1235) (Agarose)"
Met, a high affinity tyrosine kinase receptor for hepatocyte growth factor (HGF, also known as scatter factor) is a disulfide-linked heterodimer made of 45kD alpha and 145kD beta-subunits. The alpha-subunit and the amino-terminal region of the beta-subunit form the extracellular domain. The remainder of the beta-chain spans the plasma membrane and contains a cytoplasmic region with tyrosine kinase activity. Interaction of Met with HGF results in auto-phosphorylation at multiple tyrosines, which recruit several downstream signaling components, including Gab1, c-Cbl and PI3 kinase. These fundamental events are important for all of the biological functions involving Met kinase activity. The addition of a phosphate at cytoplasmic Tyr1003 is essential for Met protein ubiquitination and degradation. Phosphorylation of Tyr1234/1235 in the Met kinase domain is critical to kinase activation. Phosphorylation of Tyr1349 in the Met cytoplasmic domain provides a direct binding site for Gab1. Altered Met levels and/or tyrosine kinase activities are found in several types of tumors, including renal, colon and breast. Thus, Met is an attractive cancer therapeutic and diagnostic target. Applications: Suitable for use in Immunoprecipitation. Other applications not tested. Recommended Dilutions: Immunoprecipitation: Add 10ul of M3007-22C (well-vortexed) to 200ul of cell lysate at 1mg/ml in 1X Cell Lysis Buffer (20mM Tris, pH 7.5, 150mM sodium chloride, 1mM EDTA, 1mM EGTA, 1% Triton X-100, 2.5mM sodium pyrophosphate, 1mM beta-glycerophosphate, 1mM Na3VO4, 1ug/ml Leupeptin, 1mM PMSF., Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
