eIF2a is a 36kD protein which is ubiquitously expressed in many cell types. The eIF2 protein, which is composed of three subunits (a, b and g), is one of the key molecules in the initiation of translation. In mammalian cells, stress induces eIF2a to be phosphorylated at serine 52 by at least two kinases: the haem-controlled repressor (HCR) and the interferon inducible double stranded RNA-dependent protein kinase (PKR). Phosphorylation of eIF2a blocks the GDP-GTP exchange activity of eIF2b, leading to inhibition of protein synthesis, growth suppression, and apoptosis induction. The phosphorylation of eIF2a is an important regulatory process in protein synthesis. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1:1000, Optimal dilution determined by the researcher. Positive Control: 3T3-L1 adipocytes +/- Leukemia Inhibitory Factor (LIF). Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
