Narsoplimab (Anti-MASP2 Antibody)

Narsoplimab, also known as OMS 721, is a high-affinity fully human immunoglobulin gamma 4 (IgG4) monoclonal antibody that binds to MASP2 (Mannan-binding lectin serine protease 2) and blocks lectin pathway activation. Narsoplimab can be used in research of hematopoietic stem-cell (HSC) transplantation and SARS-CoV-2. MASP2 (Mannan-binding lectin serine protease 2) is a serine protease involved in the activation of the lectin pathway (LP) of the complement system. The recognition of mannose residues present in pathogens by MBL triggers the activation of MASP2, and the cleavage of components C4 and C2. Activation of LP is one of the main contributions in IgAN (immunoglobulin A nephropathy), a disorder that can lead to end-stage renal damage. Therapies targeting MASP2 are being developed, such as a first -in-class N-acetylgalactosamine-conjugated small interfering siRNA. FB7013, one such therapeutic, has show to inhibit LP activation during IgAN. Another strategy involves the use of recombinant antibodies, such as Narsoplimab, also known as Yartemlea, approved for the treatment of infections.
