IL-5 is a homodimeric, disulphide-linked protein produced by T-cells. Monomeric human IL-5 is a 126 amino acid protein with a reported molecular weight of 26 kD for the homodimeric protein. Mouse and human IL-5 are approximately 70% identical. IL-5 has been shown to promote the growth of immature hematopoietic BFU-E progenitors, stimulate the activation and differentiation of eosinophils, and promote the generation of cytotoxic lymphocytes. Mouse IL-5 induces the differentiation and proliferation of pre-activated B-cells and stimulates the production and secretion of IgM and IgA by B-cells stimulated with bacterial endotoxin. The TRFK5 antibody can neutralize the bioactivity of natural or recombinant IL-5. Protein function: Homodimeric cytokine expressed predominantly by T-lymphocytes and NK cells that plays an important role in the survival, differentiation, and chemotaxis of eosinophils (PubMed:2653458, PubMed:9010276). Acts also on activated and resting B-cells to induce immunoglobulin production, growth, and differentiation. Mechanistically, exerts its biological effects through a receptor composed of IL5RA subunit and the cytokine receptor common subunit beta/CSF2RB (PubMed:1495999, PubMed:22528658). Binding to the receptor leads to activation of various kinases including LYN, SYK and JAK2 and thereby propagates signals through the RAS-MAPK and JAK-STAT5 pathways respectively (PubMed:7613138).
