Human MCPinduced protein (MCPIP), also known as ZC3H12A, is an ~66kD intracellular protein that contains one RNAse domain (aa134-290), one zinc finger (aa301-324), and a prolinerich region (aa458536). Within aa426-599, human MCPIP shares ~79% aa sequence identity with mouse and rat MCPIP. Its expression is induced by inflammatory stimulation and cellular stress. It acts to dampen inflammatory responses by promoting the degradation of proinflammatory cytokine mRNAs, inhibiting NFkB activation, and antagonizing TLR signaling. MCPIP exhibits deubiquitinase activity and inhibits the biogenesis of miRNA. It also enhances inflammationinduced angiogenesis, osteocyte and adipocyte differentiation, and the cell death of cardiac myocytes. Applications: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilutions: Western Blot: 1ug/ml using untreated HeLa human cervical epithelial carcinoma cell line and RAW 264.7 mouse, monocyte/macrophage cell line and THP-1 human acute monocytic leukemia cell line, both treated with LPS, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
