Vesicle-associated membrane protein (VAMP)-associated protein B (VAP-B, also ALS8) is a 33kD, ubiquitously expressed, type IV transmembrane protein and member of the VAP family of proteins. VAP-B may play a role in vesicle trafficking. It is found in plasma and intracellular vesicle membranes as a homodimer and heterodimer with VAP-A, and interacts with VAMP1 and VAMP2. Defects in VAP-B are a cause of amyotrophic lateral sclerosis type 8 and spinal muscular atrophy autosomal dominant Finkel type. Human VAP-B is 243aa in length and contains a 222aa cytoplasmic domain plus a 21aa C-terminal intramembrane segment. Within the cytoplasmic domain is a major sperm protein (MSP) domain (aa8-111). A second isoform, named VAP-C, has a 29aa substitution for aa71-99 plus a deletion of residues corresponding to aa 100-243 in VAP-B. Over aa1-132, human VAP-B is 96% identical to mouse VAP-B. Applications: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1ug/ml, ELISA: 0.5-1ug/ml, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS, sterile 40-50% glycerol, aliquot and store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
