TINAGL1 (Tubulointerstitial nephritis antigen-like 1, also TIN-ag-RP, AZ-1 and LCN7) is a 54-57kD glycoprotein member of the peptidase C1 family of molecules. It is secreted by renal tubule epithelium, zona glomerulosa cells, vascular smooth muscle and striated muscle cells. TINAGL1 may play a role in the spatial and temporal development of the steroid component of the adrenal, in part by suppressing the expression of CYP11B1. Human TINAGL1 is synthesized as a 467 amino acid (aa) precursor. It contains a 21 a signal sequence and a 446aa mature region (aa22-467). Within the mature region is an SMB (somatomedin B) domain (aa50-98), one vWFC domain (aa105-140), and a nonenzymatic peptidase C1A region (aa204-455). There are multiple potential isoform variants. One contains an alternative start site at aa106, another shows a deletion of aa126-156, and a third possesses a 60 aa substitution for aa1-194, coupled to a 38aa insertion after aa286, and a 16aa substitution for aa365-467. Undefined forms of 40-42kD and 22kD have been noted in TINAGL1­transfected cell supernatant subjected to SDS­PAGE. Over aa22-464, human TINAGL1 shares 90% aa identity with mouse TINAGL1. Applications: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 0.5ug/ml, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
