Anti-STAT1a (Signal Transducers and Activators of Transcription 1alpha)

STATs (signal transducers and activators of transcription) are a family of cytoplasmic latent transcription factors that are activated to regulate gene expression in response to a large number of extracellular signaling polypeptides including cytokines, interferons, and growth factors. After phosphorylation by JAK tyrosine kinases, STATs enter the nucleus to regulate transcription of many different genes. Among the seven STATs (Stat1, Stat2, Stat3, Stat4, Stat5a, Stat5b, and Stat6), Stat1, Stat3, Stat5a, and Stat5b have a wide activation profile (1,2). STAT1 is activated by many different ligands including IFN family (IFN-a, IFN-b, IFN-g, and IL-10), gp130 family (IL-6, IL-11, LIF, CNTF, and G-CSF), and receptor tyrosine kinases (EGF, PDGF, and CSF-1) (3). STAT1 has two forms, the 91kD STAT1a and the 84kD STAT1b, which are encoded by the same gene with splicing variant (4). This polyclonal antibody can be used for Western blot analysis of STAT1a expression at 1:1000 to 1:2000 and for immunoprecipitation at 2 to 4 ug per sample. The antibody is human and mouse reactive. Whole cell lysate from HeLa, Jurkat, or A431 cells can be used as positive control and a 91kD band can be detected. No reaction to the 84kD STAT1b. Applications: Suitable for use in Western Blot, Immunoprecipitation. Other applications not tested. Recommended Dilution: Western Blot: 1:1000-1:2000 Immunoprecipitation: 2-4ug, Optimal dilutions to be determined by the researcher. Positive Control: Whole cell lysate from HeLa, Jurkat, or A431 cells , Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
