Product information "Anti-SHP2 (SH2 Domain-containing Protein Tyrosine Phosphatase 2, SHP-2, BPTP3, BPTP-3, CFC, MGC14433"
The steady state of protein tyrosyl phosphorylation in cells is regulated by the opposing action of tyrosine kinases and protein tyrosine phosphatases (PTPs). Several groups have independently identified a non-transmembrane PTP, designated SH-PTP1 (also known as PTP1C, HCP and SHP), which is primarily expressed in hematopoietic cells and characterized by the presence of two SH2 domains N- terminal to the PTP domain. A second and much more widely expressed PTP with SH2 domains, SH-PTP2 (also designated PTP1 D and Syp), has been identified. SHP2, a NT2 non-receptor protein tyrosine phosphatases, contains two tandem SRC homology (SH2) domains at the N-terminus of the catalytic domain. Based on crystallographic studies, the SH2 domain is believed to function as a conformational switch which can bind and inhibit the phosphatase or bind phosphoprotein and activate the enzyme. SHP2 is thought to link growth factor receptors and other signalling proteins via protein-tyrosine phosphorylation. It may play a role in mammalian budding morphogenesis and immune response. Missense mutations in the SHP2 gene have been linked to Noonan Syndrome. Additionally, SHP2 is present in diverse neurons and is thought to play a role in abroad spectrum of neuronal responses, including neuronal survival. Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Immunohistochemistry (Formalin fixed paraffin embedded): 1:50 , Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
