Anti-S100A8 (MRP-8)

Mouse S100A8 (also CP-10, calgranulin A and MRP8) is a 10kD member of the S100 family of calcium-binding proteins. The S100A8 protein is 89 amino acids (aa) in length and contains short, sequential modules. There is an N-terminal a-helix, followed by a calcium-binding EF-hand segment, and a C-terminal a-helix. S100A8 will noncovalently heterodimerize with S100A9. In the presence of calcium, this heterodimer will form a heterotetramer. The dimeric complex is both intracellular and extracellular. It binds heparan sulfate and is chemotactic for PMNs and macrophages. Mouse S100A8 is 80% and 57% aa identical to rat and human S100A8, respectively. Applications: Suitable for use in Direct ELISA, Western Blot and Immunocytochemistry. Other applications not tested. Recommended Dilutions: Western Blot: 2ug/ml using mouse leukocyte, Immunocytochemistry: 8-25ug/ml using immersion fixed NIH-3T3 mouse embryonic fibroblast cell line, Optimal dilutions to be determined by the researcher. Recommended Secondary Antibodies: I1904-74F: IgG, Murine X-Adsorbed (FITC) Pab Gt x Rt, Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
