Anti-PIWIL2 (Piwi-like Protein 2, Cancer/Testis Antigen 80, CT80, HILI)

PIWIL2 is a 100-110kD cytosolic member of the PIWI subfamily, Argonaute family of proteins. It is expressed in the germ line, particularly in premeiotic and early meiotic germ cells, plus mesenchymal stem cells, and in select cancer stem cells such as CD44+CD24- breast cancer cells. PIWIL2 likely silences mobile genetic elements in germ cells, and appears to depress stem cell proliferation by inhibiting the expression of cell cycle genes. Human PIWIL2 is 973aa in length. It contains one PAZ domain aa384-496 and a C-terminal PIWI domain aa668-959 that interact with RNA. There are multiple methylation sites on arginines. Two potential splice forms are reported, one that shows a deletion of aa887-922, and a second that contains a 12aa substitution for aa486-801. Over aa216-379, human PIWIL2 shares 96aa identity with rat PIWIL2. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1ug/ml, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
