Parathyroid Hormone (PTH) is a hormone produced by the parathyroid gland responsible for the regulation of calcium and phosphorus concentrations in extracellular fluid. Human parathyroid hormone is produced in the parathyroid gland as a 115aa single chain polypeptide, bearing a 25aa signal peptide, a 6aa pro-peptide sequence and an 84aa mature hormone. Defects in the PTH gene are a cause of familial isolated hypoparathyroidism (FIH), a condition characterized by hypocalcemia and hyperphosphatemia owing to levels of parathyroid hormone being insufficient to maintain extracellular calcium concentrations within normal parameters. Clinical features of FIH include cramps, seizures and tetany. More rarely, hypoparathyroidism like disease, Pseudohypothyroidism (PHP1A) may be caused by mutations in the GNAS gene or defects in the PTHR1 gene leading to Jansen metaphyseal chondrodysplasia (JMC), conditions where end organ resistance to PTH function exists. Applications: Suitable for use in ELISA and Immunohistochemistry. Other applications have not been tested. Recommended Dilutions: Immunohistochemistry: Frozen and paraffin sections, ELISA: 1:2000-1:20,000, Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
