Origin recognition complex subunit 1 (ORC1L) is a member of the highly conserved six subunits of the Origin Recognition Complex (ORC) which is essential for the initiation of DNA replication in eukaryotic cells. ORC complex binds specifically to origins of replication and serves as a platform for further assembly of additional transcription initiation factors such as Cdc6 and Mcm proteins. ORC1L is the largest encoded protein of the complex. Levels of this protein vary during the cell cycle, which has been shown to be controlled by Ubiquitin-mediated proteolysis after initiation of DNA replication. This protein is found to be selectively phosphorylated during mitosis. It is also reported to interact with MYST histone acetyltransferase 2 (MyST2/HBO1), a protein involved in control of transcription silencing. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1-3ug/ml, Optimal dilutions to be determined by the researcher. Positive Control: Daudi and NIH 3T3 cell lysate, Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
