Product information "Anti-Neurofilament H, 200kD (NF-H), clone 4A75"
Like most other intermediate filament proteins (IFPs), the expression of the different neuronal IFPs is both tissue-specific and developmentally regulated. The neurofilament (NF) triplet proteins (70, 160, and 200kD) occur in both the central and peripheral nervous system and are normally restricted to neurons. The 70kD NF-protein can self-assemble into a filamentous structure, whereas the 160kD and 200kD NF-proteins require the presence of the 70kD NF-protein to co-assemble. All three NF proteins can be detected by immunohistochemical methods at day 9 or 10 after gestation in the mouse embryo. The 200kD neurofilament protein is abundantly expressed in the spinal cord, cerebellum, pons and medulla. These are predominantly brain regions giving rise to long-axon neurons. Although IFPs of the neurofilament type are normally restricted to neurons, there are reports on their expression in non-neuronal cells as well. For example, in heart conduction myocytes are NF proteins expressed together with desmin. In tumorpathology ganglioneuro-blastomas and some of the neuroblastomas are strongly positive with the neurofilament antisera. In cell cultures of neural tissues the neurofilament antibodies can monitor in vitro differentiation [1]. , Applications:, Suitable for use in Immunohistochemistry (frozen and paraffin sections) and Western Blotting. Other applications not tested. Optimum dilutions to be determined by researcher. , Storage and Stability:, May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
