Product information "Anti-Nephrin"
Nephrin, also known as renal glomerulus-specific cell adhesion receptor and nephrosis 1 (NPHS1) is a 185-200 kD type I transmembrane protein belonging to the immunoglobulin (Ig) superfamily. It is expressed on podocytes and is an essential component of the interpodocyte-spanning slit diaphragm complex. Nephrin forms cis-hetero-oligomeric complexes with Neph1, followed by trans-homophilic interaction with Nephrin on opposing cells. Mutations in the Nephrin gene is the pathogenic cause of congenital nephrotic syndrome. Mature human Nephrin contains a 1033 aa extracellular region and a 165 aa cytoplasmic tail. One potential soluble splice form is known where aa's 1056-1095 are deleted, eliminating the transmembrane region. Applications: Suitable for use in ELISA, Western Blot, Immunocytochemistry and Immunohistochemistry. Other applications not tested. Recommended Dilution: ELISA: 0.5-1ug/ml. The detection limit is ~0.3ng/well., Western Blot: 0.1-0.2ug/ml. The detection limit is ~25ng/lane under non-reducing and reducing conditions. , Immunocytochemistry: 2-5ug/ml, Immunohistochemistry: 2-5ug/ml, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute by adding sterile PBS. Aliquot and store at -20°C. Reconstituted product is stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
