Monkeypox is a zoonotic disease caused by monkeypox virus (MPXV), which is a member of orthopoxvirus genus. The MPXV E8L protein is the homologous protein of vaccinia virus D8L, D8L protein binds to chondroitin sulfate (CS) on cells, revealing a different type of virus-GAG interaction.The D8L protein has also been shown to elicit strong humoral and cellular immune responses in mice. This abundant 32-kDa IMV protein plays an important role in virus entry and adsorption due to its ability to bind to chondroitin sulfate on host cells. In vitro, antibodies directed against D8L have IMV-neutralizing activity, and soluble D8L protein inhibits the adsorption of wild-type vaccinia virus to cells. In vivo, D8L is immunogenic, and immunization of mice with a DNA vaccine encoding D8L provides partial protection from lethal vaccinia virus challenge. A study by Sakhatskyy has confirmed that D8L induces strong protective antibody responses in vivo and improves the efficacy of polyvalent poxvirus DNA vaccines composed of A27L and B5R or of A27L, B5R, L1R, and A33R.
