Matriptase is an epithelial-derived, integral membrane serine protease. This protease forms a complex with the Kunitz-type serine protease inhibitor, HAI-1, and is found to be activated by sphingosine 1-phosphate. This protease has been shown to cleave and activate hepatocyte growth factor/scattering factor, and urokinase plasminogen activator, which suggest the function of this protease as an epithelial membrane activator for other proteases and latent growth factors. The expression of this protease has been associated with breast, colon, prostate, and ovarian tumors, which implicates its role in cancer invasion, and metastasis. Applications: Suitable for use in Flow Cytometry, ELISA and Western Blot. Other applications not tested. Recommended Dilution: Flow Cytometry: 2.5ug labels 10e6 cells. Validated with PC-3 human prostate cancer cell line fixed with paraformaldehyde and permeabilized with saponin , Western Blot: 0.1ug/ml, Optimal dilutions to be determined by the researcher. Recommended Secondary Antibodies: I1904-59A: IgG, H&L (FITC) Pab Rb x Sh, I1904-59F: IgG, H&L, X-Adsorbed (FITC) Pab Rb x Sh, I1904-62P: IgG, H&L, X-adsorbed (FITC) Pab Rb x Sh, I1904-62A: IgG, H&L (FITC) Pab Dk x Sh, Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
