Mucosal addressin cell adhesion molecule-1 (MadCAM-1) is a cell-surface immunogloblin (Ig) superfamily member composed of two extracellular Ig domains, followed by a mucin-like domain, a transmembrane domain, and a short cutoplasmic domain. The molecule is similar to ICAM1 and VCAM1. It interacts via its N-terminal Ig domain with the lymphocyte homing receptor alpha4 beta73, which plays a critical role in forming the gut-associated lymphoid system. MAdCAM-1 promotes the adhesion of T and B cells, monocytes/macrophages, and potentially eosinophils, basophils, and differentiated mast cells to the vascular endothelium. At least seven alternatively spliced transcripts encoding different protein isoforms have been found for this gene. The molecular weight is about 43kD, and it is highly expressed on high endothelial venules(VEC) and lamina propia venules found in the small intestine. Applications: Suitable for use in Flow Cytometry, ELISA, Immunohistochemistry and Western Blot. Other applications not tested. Recommended Dilution: Flow Cytometry: 1:10- 1:100. Use10ul of the suggested working dilution to label 10e6 cells in 100ul., Immunohistochemistry (paraffin): 1:50. Does not require protein digestion pre-treatment of paraffin embedded sections prior to staining. Does not require antigen retrieval prior to staining of paraffin sections., Western Blot: Detects a band of approximately 60kD under non-reducing conditions., Optimal dilutions to be determined by the researcher. Hybridoma: P3X63-Ag8.653 myeloma cells with spleen cells from Balb/c mice. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
