Anti-Leucyl-cystinyl Aminopeptidase/LNPEP

LNPEP (also known as PLAP, CAP, IRAP, and Oxytocinase) is a 165-175kD member of the oxytocinase subfamily, M1 family of zinc metallopeptidases. It is widely expressed after cell stimulation and is found on endothelial cells, trophoblasts, skeletal muscle cells, neurons, adipocytes, and renal distal tubule epithelium. LNPEP cleaves vasopressin and oxytocin and serves as a receptor for AngIV. Human LNPEP is a 1025aa type II transmembrane glycoprotein., It contains a 110aa cytoplasmic region (aa1110)plus an 894aa extracellular domain (aa1321025) that contains a peptidase M1 region(aa168552).There are multiple potential splice variants. There are two alternative start sites at Met15 and Met20, and the cytoplasmic tail undergoes phosphorylation at Tyr70 and Ser80. LNPEP is reportedly forms homodimers. Proteolytic cleavage between Phe154Ala155 generates a 150155kD soluble form. Over (aa 133552), human LNPEP shares 89% aa sequence identity with mouse LNPEP. Applications: Suitable for use in Western Blot and Immunoprecipitation. Other applications not tested. Recommended Dilution: Western Blot: 1ug/ml, Immunoprecipitation: 25ug/ml, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
