Product information "Anti-Ki-67 Rabbit (Clone RM521)"
Ki-67 is a non-histone nuclear and nucleolar protein and marker of cell proliferation encoded by MKI67 in humans. It is composed of an N-terminal forkhead-associated domain, a protein phosphatase 1-binding domain, a central conserved domain, 16 concatenated direct repeats, and six C-terminal leucine- and arginine-rich motif repeats. Ki-67 expression and localization is cell cycle-specific. It is localized to the perinuclear region during G1, the fibrillary component of the nucleolus during interphase, and the perichromosomal layer during chromatin condensation, with Ki-67 levels increasing from the late G1 phase to S phase and peaking at mitosis. Ki-67 is associated with proliferative activity of intrinsic cell populations in tumors and is commonly used as a marker of tumor aggressiveness in patients with breast, lung, prostate, cervical, or CNS cancers. Ki-67 is also commonly used as a marker of adult neurogenesis. Cayman's Ki-67 Rabbit Monoclonal Antibody (Clone RM521) can be used for immunohistochemistry (IHC) and Western blot (WB) applications.. Cross reactivity: (+) Ki-67: . (+) Human: .. Protein function: Required to maintain individual mitotic chromosomes dispersed in the cytoplasm following nuclear envelope disassembly (PubMed:27362226). Associates with the surface of the mitotic chromosome, the perichromosomal layer, and covers a substantial fraction of the chromosome surface (PubMed:27362226). Prevents chromosomes from collapsing into a single chromatin mass by forming a steric and electrostatic charge barrier: the protein has a high net electrical charge and acts as a surfactant, dispersing chromosomes and enabling independent chromosome motility (PubMed:27362226). Binds DNA, with a preference for supercoiled DNA and AT-rich DNA (PubMed:10878551). Does not contribute to the internal structure of mitotic chromosomes. May play a role in chromatin organization (PubMed:24867636). It is however unclear whether it plays a direct role in chromatin organization or whether it is an indirect consequence of its function in maintaining mitotic chromosomes dispersed (Probable).
