Jagged 1 is a 180kD type I transmembrane glycoprotein and member of the Delta-Serrate-Lag-2 (DSL) family of ligands that activate LIN12/Notch proteins. Human Jagged 1 is synthesized as a 1218aa precursor that contains a 33aa signal sequence, a 1034aa extracellular domain (ECD), a 26aa transmembrane segment, and a 125aa cytoplasmic region. The ECD contains a DSL domain (aa185-229), a cysteine-rich region, 15 EGF-like repeats, of which many bind calcium, and nine potential sites for N-linked glycosylation. Mature human Jagged 1 shares 97-96aa identity with mature mouse and rat Jagged 1, respectively. Jagged 1 is widely expressed in adult and fetal tissues. Jagged-Notch signaling specifies cell fate, regulates pattern formation, defines boundaries between different cell types, and modulates cell proliferation and differentiation, especially during hematopoiesis, myogenesis, neurogenesis, and development of vasculature. Mutations in human Jagged 1 are the cause of Alagille syndrome, an autosomal-dominant disorder characterized by intrahepatic cholestasis and abnormalities of heart, eye, vertebrae, as well as characteristic facial appearance. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilutions: Flow Cytometry: 2.5ug labels 10e6 cells. Using K562 human chronic myelogenous leukemia cell line, bEnd. mouse endothelioma cell line and rat cortical stem cells., Optimal dilutions to be determined by the researcher. Hybridoma: NSO myeloma cells with B cells from Balb/c mice. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C after receipt. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
