Interleukin­22 (IL­22), also known as IL­10­related T cell­derived inducible factor (IL­TIF) was initially identified as a gene induced by IL­9 in mouse T cells and mast cells. Human IL­22 cDNA encodes a 179 aa residue protein with a putative 33 aa signal peptide that is cleaved to generate a 147 aa mature protein that shares approximately 79% and 22% aa sequence identity with mouse IL­22 and human IL­10, respectively. The human IL­22 gene is localized to chromosome 12q15. Although it exists as a single copy gene in human and in many mouse strains, the mouse IL­22 gene is duplicated in some mouse strains including C57B1/6, FVB and 129. The two mouse genes designated IL­TIF-alpha and IL­TIF-beta, share greater than 98% sequence homology in their coding region. IL­22 has been shown to activate STAT­1 and STAT­3 in several hepatoma cell lines and upregulate the production of acute phase proteins. IL­22 is produced by normal T cells upon anti­CD3 stimulation in humans. Mouse IL­22 expression is also induced in various organs upon lipopolysaccharide injection, suggesting that IL­22 may be involved in inflammatory responses. The functional IL­22 receptor complex consists of two receptor subunits, IL­22R (previously an orphan receptor named CRF2­9) and IL­10 R-beta (previously known as CRF2­4), belonging to the class II cytokine receptor family. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilutions: Western Blot: 0.1ug/ml, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
