Product information "Anti-Interleukin 17F (IL-17F)"
The Interleukin 17 family proteins, comprising six members (IL-17A through IL-17F), are secreted, structurally related proteins that share a conserved cystine knot fold near the Cterminus, but have considerable sequence divergence at the Nterminus. With the exception of IL17B, all IL17 family members are disulfidelinked dimers. IL17 family proteins are pro-inflammatory cytokines that induce local cytokine production and are involved in the regulation of immune functions. Human IL-17F cDNA encodes a 163aa protein with a putative 30aa signal peptide. Among IL17 family members, IL17F is most closely related to IL17A (~44% aa sequence homology), but shares only limited sequence homology (16-30%) with IL17B, C, D and E. Human and mouse IL17F share 55% sequence identity. IL17F is expressed in activated CD4+ Tcells and activated monocytes. Although the ligands for IL17 RD and E are not known yet, it is reported that IL17 RA binds IL17A, and IL17 RB binds IL17B and IL17E. IL17 RC binds IL17A and IL17F with similarly high affinity and functions as a receptor for both IL17A and IL17F (5, 6). IL17F stimulates production of IL6, IL8, GCSF and regulates cartilage matrix turnover by increasing matrix release and inhibiting new matrix synthesis. IL17F also inhibits angiogenesis and induces production of IL2, TGFb, and monocyte chemoattractant protein1 in endothelial cells. Applications: Suitable for use in ELISA, Western Blot and Immunocytochemistry. Other applications not tested. Recommended Dilution: ELISA (Capture): 0.2-0.8ug/ml, Western Blot: 0.1-0.2ug/ml The detection limit is ~5ng/lane under non-reducing and reducing conditions., Immunocytochemistry: 15ug/ml , Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
