IGF­I receptor is a disulfide­linked heterotetrameric transmembrane protein consisting of two a and two b subunits. Both the a and b subunits are encoded within a single receptor precursor cDNA. The proreceptor polypeptide is proteolytically cleaved and disulfide­linked to yield the mature heterotetrameric receptor. The a subunit of IGF­I receptor is extracellular while the b subunit has an extracellular domain, a transmembrane domain and a cytoplasmic tyrosine kinase domain. The IGF­I receptor is highly expressed in all cell types and tissues. Essentially all of the biological activities of IGF­I and II have been shown to be mediated via IGF­I R. Applications: Suitable for use in Western Blot, Immunohistochemistry, Neutralization and Immunocytochemistry. Other applications not tested. Recommended Dilution: Western Blot: 0.1mg/ml, Immunohistochemistry (paraffin): 5-15mg/ml using immersion fixed paraffin-embedded sections of human placenta (chorionic villi) and immersion fixed frozen sections of mouse embryo, Neutralization: 0.5-1.5mg/ml in the presence of 6ng/ml recombinant human IGF-I., Immunocytochemistry:1-15ug/ml using immersion fixed MCF-7 human breast cancer cell line., Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
