Nuclear Factor-kappaB (NF-kappaB) is sequestered in the cytoplasm by the IkappaB family of inhibitory proteins that mask the nuclear localization signal of NF-kappaB, thereby preventing translocation of NF-kappaB to the nucleus. External stimuli such as tumor necrosis factor or other cytokines result in phosphorylation and degradation of IkappaB, releasing NF-kappaB dimers. NF-kappaB dimers subsequently translocate to the nucleus and activate target genes. Synthesis of IkappaBalpha is autoregulated. IkappaB proteins are phosphorylated by IkappaB kinase complex consisting of at least three proteins, IKK1/alpha, IKK2/beta, and IKK3/gamma. IKKgamma preferentially interacts with IKKbeta and is required for activation of IKK complex. IKKgamma is also known as NF-kappaB essential modulator (NEMO). The human T cell leukemia virus type I Tax oncoprotein that activates NF-kappaB binds neither to IKKalpha nor IKKbeta, but complexes directly with IKKgamma. This suggests that IKKgamma may be a key molecule acting as an adapter for oncoprotein specific signaling to IKKalpha and IKKbeta.Synonyms: IKK3, NEMO, NF-kappaB Essential Modulator.
