Product information "Anti-Histone H3.3G34R Rabbit - Biotinylated ("
Histone H3 is a nuclear protein and component of the nucleosome core, a basic unit of chromatin, that is essential for organizing genomic DNA in eukaryotic nuclei. It is a globular protein that contains an unstructured N-terminal tail that extends outside of the nucleosome core and is subject to various post-translational modifications (PTMs). Metazoans have three histone variants: canonical histone H3. DNA replication-independent histone H3. and the centromeric variant CENP-A. Histone H3. is incorporated into chromatin in a DNA replication-independent manner at sites of active transcription. A missense mutation replacing guanine with thymine in H3F3A, the gene encoding histone H3. leads to a glycine-to-arginine substitution, histone H3.3G34R, which is associated with glioblastoma multiforme in children and young adults. Cayman's Histone H3.3G34R Rabbit Monoclonal Antibody (Clone RM240) - Biotinylated can be used for chromatin immunoprecipitation (ChIP), ELISA, immunohistochemistry (IHC), and Western blot (WB) applications.. Cross reactivity: (+) Histone H3.3G34R: (-) Wild-type histone H3.3. (+) Species independent: .. Protein function: Variant histone H3 which replaces conventional H3 in a wide range of nucleosomes in active genes. Constitutes the predominant form of histone H3 in non-dividing cells and is incorporated into chromatin independently of DNA synthesis. Deposited at sites of nucleosomal displacement throughout transcribed genes, suggesting that it represents an epigenetic imprint of transcriptionally active chromatin. Nucleosomes wrap and compact DNA into chromatin, limiting DNA accessibility to the cellular machineries which require DNA as a template. Histones thereby play a central role in transcription regulation, DNA repair, DNA replication and chromosomal stability. DNA accessibility is regulated via a complex set of post-translational modifications of histones, also called histone code, and nucleosome remodeling. [The UniProt Consortium]
