HIPK3, a mammalian homolog of YAK1, is a novel 130kD nuclear co-factor that belongs to the HIPK protein family. Members of this family localize to nuclear speckles and consist of an evolutionarily conserved protein kinase domain and a homeoprotein-interacting domain. Physiological functions of HIPK3 have been predicted in different cell signaling pathways including transcriptional co-repression, cell differentiation, growth suppression, steroidogenesis and apoptosis. Research suggests that HIPK3 is a FADD-interacting protein kinase found to phosphorylate FADD and promote its binding to Fas, thereby inhibiting Fas-mediated Jun NH (2)-terminal kinase activation. HIPK3 also has been found to phosphorylate JNK and c-JUN, thus enhancing SF-1 activity for CYP11A1 transcription. HIPK3 plays a key role in cAMP-dependent signaling pathway. Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 0.5-2ug/ml, Immunohistochemistry (formalin fixed paraffin embedded): 5-10ug/ml, Optimal dilutions to be determined by the researcher. Positive Control: Testis, Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
