Heme oxygenase (HO) is a microsomal enzyme that oxidatively cleaves heme, a pro-oxidant, into carbon monoxide and biliverdin. Carbon monoxide is suggested to be a neurotransmittor and biliverdin is reduced to the antioxidant, bilirubin. Two heme oxygenase isozymes, termed HO-1 and HO-2, have been identified in rat, rabbit, monkey, and human tissues. At the amino acid level, HO-1 and HO-2 are 42% homologous. However, each isozyme is evolutionary highly conserved. On SDS-PAGE, the molecular weight of mammalian HO-1 is 31-33kD, mammalian HO-2 is 36kD. HO-1 is the inducible form of the enzyme and is therefore also referred to as stress protein Hsp32. Expression of HO-1 can be induced by a variety of stimuli such as heme, metals, hormones, UV radiation and sulfhydryl depleting agents. In contrast, HO-2 is the constituitive form of the enzyme. It is not inducible by exogeneous stimuli and HO-2 is the most prevalent form in most tissues, except the spleen where HO-1 levels are predominant under normal conditions. , , Applications: , Suitable for use in ELISA, Western Blot, Immunocytochemistry, Immunofluorescence, Immunohistochemistry, Immunoprecipitation. Other applications not tested. Recommended Dilutions: Western Blot (Colormetric): 1:1000 , Immunohistochemistry: 1:50, Immunoprecipitation (Denatured): 10ug/ml, Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
